An obligatory anaerobic bacterium was isolated from a mediator-less microbial fuel cell using starch processing wastewater as the fuel and designated as EG3. The isolate was Gram-positive, motile and rod (2.8–3.0 μm long, 0.5–0.6 μm wide). The partial 16S rRNA gene sequence and analysis of the cellular fatty acids profile suggested that EG3 clusters with Clostridium sub-phylum and exhibited the highest similarity (98%) with Clostridium butyricum. The temperature and pH optimum for growth were 37°C and 7.0, respectively. The major products of glucose and glucose/Fe(O)OH metabolism were lactate, formate, butyrate, acetate, CO2and H2. Growth was faster at the initial phase and the cell yield was higher when the medium was supplemented with Fe(O)OH than without Fe(O)OH. These results suggest that Fe(III) ion is utilised as an electron sink. Cyclic voltammetry showed that Clostridium butyricum EG3 cells were electrochemically active. The capacity of anoxygenic phototrophic bacteria to utilize aromatic hydrocarbons was investigated in enrichment cultures
with toluene. When mineral medium with toluene (provided in an inert carrier phase) was inoculated with activated sludge and
incubated under infrared illumination (> 750 nm), a red-to-brownish culture developed. Agar dilution series indicated the
dominance of two types of phototrophic bacteria. One type formed red colonies, had rod-shaped cells with budding division,
and grew on benzoate but not on toluene. The other type formed yellow-to-brown colonies, had oval cells, and utilized toluene
and benzoate. One strain of the latter type, ToP1, was studied in detail. Sequence analysis of the 16S rRNA gene and DNA-DNA
hybridization indicated an affiliation of strain ToP1 with the species Blastochloris sulfoviridis, a member of the α-subclass of Proteobacteria. However, the type strain (DSM 729) of Blc. sulfoviridis grew neither on toluene nor on benzoate. Light-dependent consumption of toluene in the presence of carbon dioxide and formation
of cell mass by strain ToP1 were demonstrated in quantitative growth experiments. Strain ToP1 is the first phototrophic bacterium
shown to utilize an aromatic hydrocarbon. In the supernatant of toluene-grown cultures and in cell-free extracts incubated
with toluene and fumarate, the formation of benzylsuccinate was detected. These findings indicate that the phototrophic bacterium
activates toluene anaerobically by the same mechanism that has been reported for denitrifying and sulfate-reducing bacteria.
The natural abundance of phototrophic bacteria with the capacity for toluene utilization was examined in freshwater habitats.
Counting series revealed that up to around 1% (1.8 × 105 cells per gram dry mass of sample) of the photoheterotrophic population cultivable with acetate grew on toluene.

AbstractBacteria from two areas in Wales have been extracted and partially identified. The areas are a copper impregnated bog in the Coed-Y-Brenin Forest with low pH and high carbon and copper content, and the Morfa Harlech salt marshes which have neutral pH and lower copper and carbon. Bacillus spp. was the predominant bacterium in both soils, but there was increased diversity of bacteria in the salt marsh soils. Bacteria from the Cu-rich, Coed-Y-Brenin soils showed higher growth densities in nutrient agar plates containing 10, 30 and 100 μg cm?3 Cu than bacteria from the salt marsh soils, indicating that bacteria from the Coed-Y-Brenin soils could tolerate higher copper concentrations. Pseudomonas spp. from both study areas grew better in media with added Cu at low levels. The possibility is discussed that the bacteria play a part in the bioavailability of Cu particularly in the copper-rich Coed-Y-Brenin area. 相似文献

Sulphidogenous microorganism communities were isolated from selected oilfield waters in the Flysch Carpathians of south-eastern Poland. Organisms were incubated using the microcosms method with application of two media: minimal medium and modified Postgate C medium with yeast extract or trisodium citrate or monocyclic hydrocarbons from the BTEX group (benzene, toluene, ethylbenzene, and xylene) as the sole carbon source. Activity of sulphidogenic, autochthonous microorganism communities was noted only on the Postgate medium. Beside active sulphate reduction – max. 70%, ca. 74% biodegradation of organic compounds was also observed in the cultures. The highest content of sulphate-reducing bacteria (SRB) in the COD (ca. 83%) was noted in cultures, in which trisodium citrate and yeast extract were applied as the sole carbon source. Molecular analysis indicated not only the presence of SRB such as Desulfobacterium autothrophicum, Desulfovibrio desulfuricans, but also other microorganisms, e.g., Geobacter metallireducens. All these taxa are obligatory or facultative anaerobes, with metabolism linked mostly with elemental sulphur and/or its oxidized forms, as well as iron. Analysis of the mineral composition of the residues confirmed the presence of elemental sulphur, testifying for the active reduction of sulphates by incompletely oxidizing sulphate reducers assigned to the SRB group. Based on the obtained results, it is concluded that the physical and chemical properties of the oilfield waters are favorable for the growth and development of sulphidogenic microorganism assemblages and mineral-forming processes conducted by them. 相似文献

We examined the Mn(II) oxidation performance of a bacterium, BY86, collected at Yunotaki Falls Hokkaido, Japan. The bacterium showed rapid oxidation of Mn(II), and brown precipitates containing Mn formed within a few days of incubation. The presence of higher oxidation states of Mn than Mn(II) was ascertained by the UV-vis and XANES sutdy. This bacterium did not oxidize As(III) to As(V) in the absence of Mn. In the presence of Mn, however, As(III) was rapidly oxidized to As(V) on the cell surfaces. These findings indicate that BY86 does not have the ability to directly oxidize As(III) to As(V) within a short period of contact, but indirectly oxidizes it by the Mn oxides generated on the cell surfaces. A phylogenetical study disclosed that BY86 was most closely related to Bacillus cereus with an identity of 99.90%. It is expected that our findings in this study will contribute to the study of Mn(II)-oxidizing bacteria, which play an important role in the biogeochemical cycling of Mn as well as other trace elements including As. 相似文献
